Service Overview

Overexpression lines fail in predictable ways — expression too high to be physiological, silencing over passage, a tag that disrupts localisation, or toxicity that selects for low expressors. Construct design addresses each of these before transfection. We design the cassette, optimise the coding sequence for the host, select promoter strength appropriate to the intended use, and specify how expression level and stability will be confirmed.

Schematic: raw reads become a quantified matrix, then tested differences, then annotated populations — with the statistics matched to the study design.

Key Features & Advantages

Promoter Strength Chosen Deliberately
Strong constitutive promoters are not always right. Where physiological expression matters, weaker or endogenous-like promoters are proposed, with the trade-off stated.
Inducible Systems Where Control Matters
For toxic or dosage-sensitive genes, inducible architectures are designed with attention to leakiness, which is the usual reason such systems disappoint.
Tag Placement Modelled
Tag terminus and linker are chosen against predicted effects on folding, secretion signals and localisation rather than by convention.
Silencing Risk Considered
Element choice and integration strategy take account of long-term expression stability, since silencing over passage is a common late failure.

Questions We Are Asked Before Scoping

Should we use the strongest promoter available?

▸Usually not. Overexpression far above physiological level produces artefacts including mislocalisation and aggregation. We match promoter strength to the question being asked.

Why did our line lose expression over time?

▸Silencing of the integrated cassette is common, particularly with certain promoters and random integration. Element choice and targeted integration reduce the risk, and we design for it.

N-terminal or C-terminal tag?

▸It depends on the protein — signal peptides, termini involved in function and localisation determinants all matter. We model both and recommend, and where it is genuinely uncertain we suggest making both.

Interpretation and Limitations

This is a computational (in silico) service. Designs and predictions prioritise hypotheses and direct experimental effort; they do not by themselves establish that an edit, construct, molecule or assay will perform as intended. Laboratory execution — editing, cloning, expression, synthesis and testing — is planned with you and carried out by your own team or a qualified partner. Deliverables are for research use only and are not intended for diagnostic or therapeutic use.

Is This the Right Service for You?

Choose this when
A stable line expressing a target is needed for assay development or screening
Complementation or rescue experiments require controlled expression
Variants must be expressed in parallel for functional comparison
Consider another service when
Expression must stay under endogenous control

What Goes Wrong, and How We Handle It

These are the failure modes that produce confident-looking results which do not survive the next experiment. Each one is designed against rather than discovered afterwards.

Expression so far above physiological level that the biology becomes artefact
Promoter strength is matched to the intended use, with the trade-off stated rather than defaulting to the strongest available.
Silencing of the cassette over passage
Element choice and integration strategy account for long-term stability, and verification over passage is written into the plan.
A tag that disrupts folding, secretion or localisation
Both termini are assessed structurally and the placement least likely to interfere is recommended.

A Worked Example

A toxic transgene that selected for low expressors

The situation
Stable pools lost expression within a few passages whenever a strong constitutive promoter was used.
What the work involved
The transgene was assessed for likely toxicity, an inducible architecture was designed with leakiness as an explicit criterion, and integration strategy was revised.
What came back
An inducible cassette with measurable background, plus a weaker constitutive alternative for comparison. Verification over ten passages was built into the delivered plan.

Illustrative of the shape this work takes. Details are generalised, no client or target is identified, and scope and outcome vary with the system in question.

Service Workflow

1
Scientific Consultation
We discuss the objective, the decision it supports and the material and data you already hold.
2
Feasibility Review
We check that the target, system and available information can support the approach, and say plainly where they cannot.
3
Technical Proposal
Scope, methods, deliverables, timeline and price are confirmed in writing, together with the limitations we expect to report.
4
Design and Analysis
The design work runs on documented tools and parameters, with quality checks and alternatives compared where that is informative.
5
Delivery and Follow-Up
You receive the designs, the supporting analysis and a validation plan, plus support interpreting the results once the experiments run.
