Service Overview

GPCRs are the most commercially important receptor family and among the least forgiving to express. Expression level changes apparent pharmacology, the chosen readout determines which pathway you see, and the host cell background contributes signalling components you may not have accounted for. We design the construct, recommend the readout that matches the biology you care about, and model the receptor where structural insight informs mutagenesis or ligand work.

Schematic: substitutions are scored at every position for stability and function, with catalytic and binding residues protected from redesign.

Key Features & Advantages

Expression Level Treated As A Variable
Receptor density changes agonist potency and apparent efficacy. Construct design targets a level appropriate to the pharmacology being measured rather than the maximum achievable.
Readout Matched To Pathway
Second messenger, arrestin recruitment and label-free readouts report different events. We recommend based on the receptor coupling and the question, and note what each readout will miss.
Structural Modelling Where It Helps
Receptor models, including active and inactive states where templates allow, support mutagenesis design, ligand docking and interpretation of unexpected pharmacology.
Host Background Considered
Endogenous receptor and signalling component expression in the chosen host is checked, since background activity is a frequent source of confusing results.

Questions We Are Asked Before Scoping

Why does our receptor not reach the surface?

▸Common causes are missing signal peptide requirements, misfolding, absent chaperones or an unsuitable host. Sequence and structural analysis usually narrows it, and construct variants are designed to test the likely causes in parallel.

Which assay readout should we use?

▸It depends on coupling and on whether pathway-biased behaviour matters. We recommend one primary and one orthogonal readout, because single-readout pharmacology is frequently misleading.

Can you model a receptor with no structure?

▸Yes, using family templates and prediction, with confidence reported per region. Binding site conclusions from such models are treated as hypotheses for mutagenesis rather than as established.

Interpretation and Limitations

This is a computational (in silico) service. Designs and predictions prioritise hypotheses and direct experimental effort; they do not by themselves establish that an edit, construct, molecule or assay will perform as intended. Laboratory execution — editing, cloning, expression, synthesis and testing — is planned with you and carried out by your own team or a qualified partner. Deliverables are for research use only and are not intended for diagnostic or therapeutic use.

Is This the Right Service for You?

Choose this when
A functional receptor line is needed for screening or compound profiling
Biased signalling across pathways is part of the question
An existing line has a poor signal window or inconsistent pharmacology
Consider another service when
A soluble target is being expressed
Gene Expression Cell Line Design
Receptor stabilisation for structural work is the goal
Membrane Protein Design

What Goes Wrong, and How We Handle It

These are the failure modes that produce confident-looking results which do not survive the next experiment. Each one is designed against rather than discovered afterwards.

Receptor density distorting apparent potency and efficacy
Expression level is treated as a design variable, targeted to the pharmacology being measured rather than maximised.
A single readout missing the pathway that matters
A primary and an orthogonal readout are recommended, with what each will miss stated explicitly.
Endogenous receptors in the host confounding the assay
Host background expression is profiled before the cell line is chosen.

A Worked Example

A receptor that would not reach the cell surface

The situation
Expression was detectable by western blot but surface staining and functional response were both negative.
What the work involved
The sequence was analysed for signal peptide requirements and trafficking motifs, the receptor was modelled, and construct variants with different signal sequences and chaperone strategies were designed.
What came back
Four constructs testing distinct trafficking hypotheses in parallel, with the assay readout and reference compounds specified so a positive result would be interpretable immediately.

Illustrative of the shape this work takes. Details are generalised, no client or target is identified, and scope and outcome vary with the system in question.

What Published Work Reports

The results below were published by independent groups and are cited to show what these computational approaches have achieved in the literature. They are not ICDMO project results, and outcomes vary substantially between targets, datasets and programmes. We do not promise comparable results for any particular project.

Around three quarters of predicted ligand poses fall within 2 Å

Current deep-learning structure prediction, evaluated on an independent protein–ligand benchmark, placed roughly 76% of ligands within 2 Å of the experimental pose, and reported approximately 80% by coordinate error — a substantial improvement over classical docking baselines on the same set, achieved without supplying a binding site. The corollary matters as much as the headline: roughly a quarter of cases still fall outside that threshold, which is why we report confidence per interface rather than delivering a single model.

Service Workflow

1
Scientific Consultation
We discuss the objective, the decision it supports and the material and data you already hold.
2
Feasibility Review
We check that the target, system and available information can support the approach, and say plainly where they cannot.
3
Technical Proposal
Scope, methods, deliverables, timeline and price are confirmed in writing, together with the limitations we expect to report.
4
Design and Analysis
The design work runs on documented tools and parameters, with quality checks and alternatives compared where that is informative.
5
Delivery and Follow-Up
You receive the designs, the supporting analysis and a validation plan, plus support interpreting the results once the experiments run.

Note: ICDMO delivers computational (in silico) services. Results prioritise hypotheses for testing; they do not by themselves establish binding, biological activity, efficacy or safety. All deliverables are for research use only and are not intended for diagnostic or therapeutic use. Downstream experimental work is scoped separately and executed through qualified partners.
